Inhibition by acetylcholine of the norepinephrine release evoked by potassium in canine saphenous veins.

In the dog's saphenous vein acetylcholine inhibits the norepinephrine release caused by nerve stimulation, but not that caused by tyramine. Experiments were performed to determine whether acetylcholine affects the release of norepinephrine evoked by high K+ concentrations. We recorded changes in isometric tension of dog saphenous vein strips. Acetylcholine (5 X 10(-9) to 10(-6) g/ml) caused dos...

Beta-amyloid-related peptides inhibit potassium-evoked acetylcholine release from rat hippocampal slices.

The 4 kDa beta-amyloid (A beta) protein, a major component of cerebral and cerebrovascular plaques in Alzheimer's disease (AD), is derived from the proteolytic cleavage of a larger, membrane-bound precursor, the A beta precursor protein (APP). Until recently, it was assumed that an aberrant AD-specific proteolysis generated A beta peptides, which subsequently could initiate and/or contribute to...

Tubocurarine and pancuronium inhibit evoked release of acetylcholine from the mouse hemidiaphragm preparation.

Using a sensitive radioactive method that measures selectively the evoked release of acetylcholine, it was demonstrated that, when stimulating at 50 Hz, tubocurarine or pancuronium 2 X 10(-5) mol litre-1 or hexamethonium 10(-3) mol litre-1 significantly decreased the evoked release of acetylcholine in the mouse in vitro phrenic nerve-hemidiaphragm preparation.

Apolipoprotein E4 reduces evoked hippocampal acetylcholine release in adult mice

Apolipoprotein E4 (apoE4) is the most prevalent genetic risk factor for Alzheimer's disease. We utilized apoE4-targeted replacement mice (approved by the Tel Aviv University Animal Care Committee) to investigate whether cholinergic dysfunction, which increases during aging and is a hallmark of Alzheimer's disease, is accentuated by apoE4. This revealed that levels of the pre-synaptic cholinergi...

Characterization of nicotinic acetylcholine receptors that modulate nicotine-evoked [3H]norepinephrine release from mouse hippocampal synaptosomes.

Nicotine's modulation of hippocampal noradrenergic neurotransmission may contribute to its mnemonic properties, but the nicotinic acetylcholine receptor (nAChR) subtypes that modulate terminal release of norepinephrine are unknown. In the present study, we used a number of subtype-selective alpha-conotoxins in combination with nicotinic receptor subunit-deficient mice to characterize nAChRs tha...
